菌種簡介：
1、菌種名(ming)稱：ATCC 9027 銅綠假單胞菌(jun)(綠膿桿菌(jun))
2、規(gui)格：斜面□ 凍干管(guan)□
二、保(bao)藏條件：凍干菌(jun)種應在2-8℃保存；斜面菌(jun)種放15-20度保存，15-20天(tian)轉接一次。
三、培養條(tiao)件：
1. 培養基：營養瓊脂培養基(ji)（NA）
胰(yi)蛋白棟大豆肉湯培養基（TSA）
LB培(pei)養基 以(yi)上培養基三選一即可
2. 培養溫度： 37℃
3. 培養時(shi)間：18- 24h
四、真空凍干菌種的(de)復(fu)蘇培養：
1、安瓿(bu)管開封：用浸過75%酒精的脫脂棉擦凈安瓿(bu)管，用火焰加熱(re)其頂(ding)端，滴少量無菌(jun)水至加熱(re)頂(ding)端使之破(po)(po)裂，用銼刀或鑷子敲(qiao)下已破(po)(po)裂的安瓿(bu)管頂(ding)端。
2、菌種恢復培養(yang)：用無(wu)菌(jun)吸(xi)(xi)管(guan)吸(xi)(xi)取(qu) 0.1-0.2毫升的培(pei)養(yang)(yang)液或(huo)無(wu)菌(jun)水，滴入安瓿管(guan)內輕輕振蕩，使凍干菌(jun)體溶解呈懸浮(fu)狀，吸(xi)(xi)取(qu)全部菌(jun)體懸浮(fu)液，移植于1-2支建議的斜面培(pei)養(yang)(yang)基上(shang)，并在建議的溫度(du)下培(pei)養(yang)(yang)。
五(wu)、注意事項：
1、微生物(wu)菌(jun)種應(ying)保藏于低溫、清潔和干燥的地(di)方(fang)，室(shi)溫放置時間過長會導致(zhi)菌(jun)種衰退；
2、菌(jun)種操作應在無菌(jun)條件下進行，防止雜菌(jun)污染(ran)；
3、凍干(gan)菌復蘇后，應根據菌種(zhong)狀況及(ji)時轉接傳代
