凍干菌種的復(fu)蘇(su)培養：
1凍(dong)干管(guan)開封：用(yong)浸過75%酒(jiu)精的脫脂(zhi)棉擦凈凍(dong)干管(guan)，用(yong)火焰加熱其頂端(duan)，滴少(shao)量無(wu)菌(jun)水至加熱頂端(duan)使之破裂(lie)，用(yong)銼刀或鑷子敲下已破裂(lie)的凍(dong)干管(guan)頂端(duan)。
2菌種恢(hui)復培(pei)(pei)(pei)養(yang)(yang)(yang)：用無(wu)菌吸管(guan) 0.3-0.5毫升適宜到液(ye)(ye)體(ti)培(pei)(pei)(pei)養(yang)(yang)(yang)基，滴入(ru)凍干管(guan)內輕輕振蕩(dang)，使凍干菌體(ti)溶解呈(cheng)懸(xuan)浮狀(zhuang)，吸取全部菌體(ti)懸(xuan)浮液(ye)(ye)，移植(zhi)于1-2支建(jian)議的斜面(mian)培(pei)(pei)(pei)養(yang)(yang)(yang)基上，并在(zai)建(jian)議的溫度(du)下培(pei)(pei)(pei)養(yang)(yang)(yang)。

1、微(wei)生物菌種應保藏于低溫、清潔和干燥的地方，室溫放置(zhi)時間過長會導致菌種衰(shuai)退；
2、菌(jun)種操作應在無菌(jun)條件下(xia)進行，防止雜菌(jun)污染；
3、斜面菌(jun)種(zhong)(zhong)保藏(zang)時間(jian)通(tong)常為3-6個月，應根據菌(jun)種(zhong)(zhong)狀況及時轉接(jie)；凍(dong)干菌(jun)種(zhong)(zhong)保藏(zang)時間(jian)通(tong)常為2-15年；
