凍(dong)干菌種的復蘇培(pei)養(yang)：
1凍(dong)干管(guan)開封(feng)：用浸過75%酒精的脫脂棉擦凈凍(dong)干管(guan)，用火焰加熱(re)其頂端(duan)，滴少量無菌水(shui)至(zhi)加熱(re)頂端(duan)使之破裂，用銼(cuo)刀或鑷子敲下已破裂的凍(dong)干管(guan)頂端(duan)。
2菌(jun)(jun)(jun)種恢復培(pei)養(yang)：用無(wu)菌(jun)(jun)(jun)吸(xi)管(guan) 0.3-0.5毫(hao)升適宜到液體培(pei)養(yang)基(ji)，滴(di)入凍干(gan)管(guan)內輕(qing)輕(qing)振蕩，使凍干(gan)菌(jun)(jun)(jun)體溶解呈懸浮狀，吸(xi)取全(quan)部(bu)菌(jun)(jun)(jun)體懸浮液，移(yi)植(zhi)于1-2支建(jian)議的(de)(de)斜面培(pei)養(yang)基(ji)上，并在建(jian)議的(de)(de)溫度下培(pei)養(yang)。

注意事項
1、微生(sheng)物(wu)菌種應(ying)保藏于(yu)低溫、清潔和干燥的地方，室溫放置時間過長會導致菌種衰退；
2、菌種操作(zuo)應(ying)在無菌條(tiao)件(jian)下進行，防止雜菌污(wu)染；
3、斜面菌(jun)種(zhong)(zhong)(zhong)保藏(zang)時(shi)間(jian)通(tong)常為(wei)3-6個月，應根(gen)據菌(jun)種(zhong)(zhong)(zhong)狀況及時(shi)轉接；凍干菌(jun)種(zhong)(zhong)(zhong)保藏(zang)時(shi)間(jian)通(tong)常為(wei)2-15年；
