凍干菌種的復蘇培養(yang)：
1凍干管開封：用浸過75%酒精的(de)脫脂棉擦(ca)凈凍(dong)干管(guan)，用(yong)火焰加熱其頂端(duan)，滴少量無菌水至(zhi)加熱頂端(duan)使之破(po)裂，用(yong)銼刀或鑷子敲下已破(po)裂的(de)凍(dong)干管(guan)頂端(duan)。
2菌種恢(hui)復培無養：用菌吸管(guan) 0.3-0.5毫升適宜到(dao)液體(ti)(ti)(ti)培(pei)養基，滴入凍(dong)干(gan)管內輕(qing)輕(qing)振蕩，使凍(dong)干(gan)菌體(ti)(ti)(ti)溶解呈(cheng)懸浮(fu)狀(zhuang)，吸取全部菌體(ti)(ti)(ti)懸浮(fu)液，移(yi)植于1-2支建議的(de)斜(xie)面培養基上，并在建議的(de)溫度下培養。

注意事項
1、微生物(wu)菌種應保(bao)藏于低溫、清(qing)潔和(he)干燥(zao)的地方(fang)，室溫放置時間過(guo)長會(hui)導致菌種衰退(tui)；
2、菌(jun)(jun)種操(cao)作(zuo)應在無菌(jun)(jun)條件下進行，防止雜菌(jun)(jun)污染；
3、斜面菌種保藏時間通常為(wei)3-6個(ge)月，應根據(ju)菌種(zhong)(zhong)狀況及時轉接；凍干菌種(zhong)(zhong)保(bao)藏時間通(tong)常為2-15年；
