凍干菌種的復蘇培養：
1凍(dong)干管(guan)開(kai)封(feng)：用(yong)浸過7。5%酒(jiu)精(jing)的脫(tuo)脂(zhi)棉擦(ca)凈凍(dong)干管(guan)，用(yong)火焰加熱其(qi)頂端(duan)，滴少量無菌水(shui)至(zhi)加熱頂端(duan)使之破裂(lie)，用(yong)銼刀或鑷(nie)子敲下已破裂(lie)的凍(dong)干管(guan)頂端(duan)
2菌種恢(hui)復培(pei)養(yang)：用無(wu)菌吸管 0.3-0.5毫(hao)升(sheng)適(shi)宜到液體(ti)培(pei)養(yang)基，滴入(ru)凍干管內輕(qing)輕(qing)振(zhen)蕩，使凍干菌體(ti)溶解呈懸浮狀，吸取全(quan)部菌體(ti)懸浮液，移植于(yu)1-2支建議的斜面培(pei)養(yang)基上，并在建議的溫度下培(pei)養(yang)。

1、微生(sheng)物菌(jun)種(zhong)應(ying)保藏(zang)于低溫、清潔和干(gan)燥(zao)的地(di)方(fang)，室溫放置時(shi)間過長(chang)會導(dao)致菌(jun)種(zhong)衰退(tui)；
2、菌(jun)種操作(zuo)應(ying)在無菌(jun)條件(jian)下進行，防止雜(za)菌(jun)污染(ran)；
3、斜面菌(jun)種(zhong)(zhong)保藏時間(jian)(jian)通常為3-6個月，應根據菌(jun)種(zhong)(zhong)狀況及時轉接；凍(dong)干(gan)菌(jun)種(zhong)(zhong)保藏時間(jian)(jian)通常為2-15年；
